Open Spin Microscopy and Functional Imaging in Zebrafish through Light Sheet Microscopy

WHEN SIZE MATTERS - Webinar 2

Speakers: Dr. Emilio J. Gualda (Microscopy Developer, IGC- Instituto Gulbenkian de Ciência, Portugal) and Kevin Frischmann (Senior Application Scientist, Bitplane).

Open Spin Microscopy

Open software and hardware approaches in microscopy have gained interest recently as a solution for many research laboratories worldwide, in order to reduce costs and simplify training procedures. We will present the open source platform for light-sheet microscopy and optical projection tomography, developed at the Instituto Gulbenkian de Ciência, based on Micromanager and Arduino.

We will describe its components, implementation solutions and operation, as well as some of the research projects that are currently been carried out using those set-ups.

Functional imaging in Zebrafish through light sheet microscopy

Recently, the ability of imaging the whole brain of zebrafish at high speed through light sheet microscopy has opened the door of new experiments on brain activity and the response to stimuli to understand behavior. In this presentation we will introduce the basic concepts behind functional imaging and review the latest advance in the technology.

We will also describe in detail the setup build in collaboration between the Instituto Gulbenkian de Ciência and the Fundaçaõ Champalimaud, pointing out some of the future challenges of this research field.

Key learning objectives:

  • How to use Open Spin Microscopy to build SPIM/DSLM/OPT system.
  • Functional Zebrafish brain imaging using lightsheet (SPIM) microscopy setup.
  • Imaris Software as an analytical tool (object detection and tracking) for long time series.
